In vitro and in vivo NIR fluorescence lifetime imaging with a time-gated SPAD camera

Résumé

Near-infrared (NIR) fluorescence lifetime imaging (FLI) provides a unique contrast mechanism to monitor biological parameters and molecular events in vivo. Single-photon avalanche diode (SPAD) cameras have been recently demonstrated in FLI microscopy (FLIM) applications, but their suitability for in vivo macroscopic FLI (MFLI) in deep tissues remains to be demonstrated. Herein, we report in vivo NIR MFLI measurement with SwissSPAD2, a large time-gated SPAD camera. We first benchmark its performance in well-controlled in vitro experiments, ranging from monitoring environmental effects on fluorescence lifetime, to quantifying Forster resonant energy transfer (FRET) between dyes. Next, we use it for in vivo studies of target-drug engagement in live and intact tumor xenografts using FRET. Information obtained with SwissSPAD2 was successfully compared to that obtained with a gated intensified charge-coupled device (ICCD) camera, using two different approaches. Our results demonstrate that SPAD cameras offer a powerful technology for in vivo preclinical applications in the NIR window. Fluorescence

La fluorescence est une émission lumineuse provoquée par l'excitation des électrons d'une molécule (ou atome), généralement par absorption d'un photon immédiatement suivie d'une émission spontanée. Fluorescence et phosphorescence sont deux formes différentes de luminescence qui diffèrent notamment par la durée de l'émission après excitation : la fluorescence cesse très rapidement tandis que la phosphorescence perdure plus longtemps. La fluorescence peut entre autres servir à caractériser un matériau.

In vivo

In vivo (en latin : « au sein du vivant ») est une expression latine qualifiant des recherches ou des examens pratiqués sur un organisme vivant, par opposition à in vitro ou ex vivo. Les essais cliniques sont une forme de recherche in vivo, en l'occurrence sur des humains. L’expérimentation animale est une des formes principale de recherche in vivo, impliquant généralement d’utiliser des animaux de diverses espèces comme modèles de fonctionnements biologiques ou de maladies ou comme bioréacteurs pour produire des virus, des anticorps ou des produits sanguins, notamment.

Fluorescence in the life sciences

Fluorescence is used in the life sciences generally as a non-destructive way of tracking or analysing biological molecules. Some proteins or small molecules in cells are naturally fluorescent, which is called intrinsic fluorescence or autofluorescence (such as NADH, tryptophan or endogenous chlorophyll, phycoerythrin or green fluorescent protein). Alternatively, specific or general proteins, nucleic acids, lipids or small molecules can be "labelled" with an extrinsic fluorophore, a fluorescent dye which can be a small molecule, protein or quantum dot.
