Résumé

Background: The control of the functional pancreatic beta-cell mass serves the key homeostatic function of releasing the right amount of insulin to keep blood sugar in the normal range. It is not fully understood though how beta-cell mass is determined.

Glycémie

La glycémie est la concentration (par extension, le taux) de glucose dans le sang, ou le plus souvent dans le plasma sanguin. Elle est mesurée en général en millimoles de glucose par litre de sang, rarement en milligrammes de glucose par décilitre de sang, ou encore en grammes de glucose par litre de sang. La régulation de la glycémie est un système de régulation complexe, mettant en œuvre des hormones (dont les deux principaux antagonistes insuline, hypoglycémiante, et glucagon, hyperglycémiant) ainsi que divers organes (pancréas, foie, rein).

Cellule bêta

Les cellules bêta (β-cells pour les anglophones) sont l'un des types cellulaires du pancréas, et plus particulièrement des îlots de Langerhans. Chez l'être humain, elles représentent 50 à 70 % des cellules de ces îlots. Ce sont elles qui synthétisent et sécrètent l'insuline et l'amyline. Chez les patients atteints de diabète de type 1, la masse et la fonction de ces cellules diminue, entraînant une sécrétion insuffisante d'insuline et une hyperglycémie.

Pancreatic beta cell function

Pancreatic beta cell function (synonyms Gβ or, if calculated from fasting concentrations of insulin and glucose, HOMA-Beta or SPINA-GBeta) is one of the preconditions of euglycaemia, i.e. normal blood sugar regulation. It is defined as insulin secretory capacity, i.e. the maximum amount of insulin to be produced by beta cells in a given unit of time. Beta cells play a paramount role in glucose homeostasis. Progressive loss of insulin secretory capacity is a key defect associated with the transition from a healthy glycaemic state to hyperglycaemia, characteristic of untreated diabetes mellitus.
