Regulation of Episomal Gene Expression by KRAB/KAP1-Mediated Histone Modifications

Résumé

KAP1 is an essential cofactor of KRAB zinc finger proteins, a family of vertebrate-specific epigenetic repressors of largely unknown functions encoded in the hundreds by the mouse and human genomes. So far, KRAB/KAP1-mediated gene regulation has been studied within the environment of chromosomal DNA. Here we demonstrate that KRAB/KAP1 regulation is fully functional within the context of episomal DNA, such as adeno-associated viral and nonintegrated lentiviral vectors, and is correlated with histone modifications typically associated with this epigenetic regulator.

Human genome

The human genome is a complete set of nucleic acid sequences for humans, encoded as DNA within the 23 chromosome pairs in cell nuclei and in a small DNA molecule found within individual mitochondria. These are usually treated separately as the nuclear genome and the mitochondrial genome. Human genomes include both protein-coding DNA sequences and various types of DNA that does not encode proteins. The latter is a diverse category that includes DNA coding for non-translated RNA, such as that for ribosomal RNA, transfer RNA, ribozymes, small nuclear RNAs, and several types of regulatory RNAs.

Histone

Les histones sont des protéines localisées dans le noyau des cellules eucaryotes et dans les archées. Elles sont les principaux constituants protéiques des chromosomes. Elles sont en effet étroitement associées à l’ADN dont elles permettent la compaction, cette action formant des structures appelées nucléosomes : l'ADN est enroulé autour des histones comme du fil autour d'une bobine. Les histones sont très riches en acides aminés basiques (lysine et arginine), dont la charge positive à pH physiologique permet une interaction forte avec les groupements phosphate de l'ADN qui portent des charges négatives.
